EBV-associated mononucleosis leads to long-term global deficit in T-cell responsiveness to IL-15
Blood Sauce et al. 108: 11

Supplemental materials for: Sauce et al

Files in this Data Supplement:

  • Figure S1. IL-7Rα and IL-15Rα expression on CMV tetramer-specific CD8+ T cells in a CMV-IM patient (JPG, 59.1 KB) -

The IL-R status of CD8+ T cells specific for a CMV-encoded epitope, TPR, was analysed (as for EBV epitope-specific responses in Figure 3) in a CMV-IM patient studied during acute disease, and again 3 and 12 months later. Results are expressed as the percentage of epitope-specific cells expressing IL-7R or IL-15R. Equivalent data for healthy CMV carriers are shown in parallel (light grey shading) and combine results from CD8 memory populations specific for TPR and for six other CMV epitopes; the horizontal line represents the median value.

  • Figure S2. IL-7Rα and IL-15Rα expression on cultured cells (JPG, 111 KB) -

PBMCs from post-IM donor IM70.6 and from a healthy EBV carrier were cultured in (A) IL-7 or (B) IL-15 at 1ng/ml plus anti-CD3/CD28 coated-beads as a stimulus as described in Figure 8. Input cells (day 0) and cells harvested on days 3 and 10 of culture were co-stained for either IL-7R or IL-15R and for CD8. Representative staining profiles from both donors are shown for (A) IL-7R expression in IL-7 exposed cultures and (B) IL-15R expression in IL-15 exposed cultures.
